With daniatapi.flt /RSJ I can have lockcdr.flt and still have my S: access to the CD/DVD without attaching it.  All this works fine with RSJ 5.06. But with any newer release I get the aforementioned error when trying to copy a DVD.

I have no DVD access with Dani 1.8.1 / 0.4.0. Currently, I'm rolled back to 1.7.10 and 0.3.18. Perhaps this is your problem? I've been using RSJ 6.0 (just upgraded to 6.01 last night, amidst all this discussion).

You probably already checked this but seeings as I got bit by it when I updated to Dani's 1.8.1/0.4.0 myself I'll ask, is there a dani driver on the root of the boot drive?  I had checked /os2 and /os2/boot but didn't even think of the root due to 1) I would usually never put a file there 2) not recalling that a basedev would read the root.
